Mingxuan Wu Group Develops New Method for Histone H4 Using Sortase-mediated Ligation

The Mingxuan Wu group at Westlake University recently published a research article titled “Deciphering histone H4 lysine acetylation and methylation via sortase-mediated semisynthesis” in Cell Report Physical Science.
In their study, they introduced a semisynthesis method for histone H4 using sortase-mediated ligation, allowing for the preparation of nucleosomes containing histone H4 with multiple modifications. This highly efficient method facilitated the identification of a novel H4K20me3 reader protein. Their findings are expected to accelerate the understanding of H4 modifications, particularly H4K20 methylation.
